1. Sinorhizobium meliloti is a soil bacteria which easily change from mucoid to non-mucoid phenotype in laboratory culture.
2. S. meliloti GR4 strain shows more frequently non-mucoid variants than AK21 and RMO17 strains.
3. ExpR gene is a good candidate to analyze the mucoid and non mucoid phenotypes.
4. The single mutations T337 and T565 present in the gene ExpR of the mutants GR4-5.1.1. and GR4-6.3.1 respectively affect deeply the ExpR gene and probably explain the non-mucoid phenotype.

Figure 1. Sinorhizobium meliloti mucoid and non-mucoid phenotypes.

Figure 2. PCR, DNA electrophoresis...

Figure 3. Electropherograms showing the point mutations found.

Figure 4. Consequences of point mutations on ExpR gene.

Hurry up!!!

  5. Figure 4. Significance of the single mutations detected on ExpR gene. Representation of the Open Reading frames corresponding to the ExpR gene (246 aa) of the wt(A), T337 mutant (B) and T565 mutant (C); note that a stop codon generates a truncated protein of 112 aa and 138 aa, respectively.
